CJC-1295 (No DAC) + Ipamorelin

Also known as: CJC-1295 no DAC + Ipamorelin blend; Mod GRF 1-29 + Ipamorelin; CJC/Ipa; GH pulse stack; 10mg blend (5/5); 20mg blend (10/10)

NOT FDA APPROVED Tier 3 – Early human Growth Hormone

Primary research focus: Growth hormone axis research (combined GHRH analog and selective ghrelin-receptor agonist)

Last updated July 2026 · Reviewed against FDA labeling and published research.

What is CJC-1295 (No DAC) + Ipamorelin?

This blend pairs CJC-1295 without DAC (Modified GRF 1-29, a short-acting GHRH analog) with Ipamorelin (a selective ghrelin-receptor agonist) in a single co-lyophilized vial. It is the most established of the growth hormone secretagogue pairings, based on the two-receptor rationale that a GHRH analog and a GH secretagogue amplify each other's effect on a single pulse. It is commonly sold as a 10 mg vial (5 mg of each) and also as a 20 mg vial (10 mg of each). It is a research blend with no FDA approval, and both components' compounding status is unresolved.

How it works

The pairing rationale is genuinely mechanistic rather than marketing, and it is the strongest of the blends on this basis: the two components act on different pituitary receptors and their effects are synergistic rather than merely additive. CJC-1295 without DAC binds the GHRH receptor on somatotrophs, raising cAMP and driving growth hormone synthesis and release; it is short-acting, with a half-life of roughly 30 minutes, producing a discrete pulse. Ipamorelin binds the growth hormone secretagogue receptor (GHS-R1a, the ghrelin receptor), driving Gq/phospholipase C signaling with calcium release and amplifying the release of stored GH, with a half-life of about 2 hours and notable selectivity: it does not meaningfully raise cortisol, prolactin, or appetite at GH-releasing doses. Combining a GHRH signal with a secretagogue signal produces a larger, more physiological GH pulse than either alone. That said, the synergy is established for the receptor classes generally, not for this specific fixed-ratio blend in a published human trial.

How it’s supplied

Not an FDA-approved or commercially manufactured medicine. It is sold by research-chemical vendors and has been prepared by compounding pharmacies during periods when that was permitted, as a single co-lyophilized vial, most commonly 10 mg total (5 mg of each) and also 20 mg total (10 mg of each). As with other blends, confirm the per-component split rather than relying on the headline vial number.

Units shown are for a U-100 insulin syringe (100 units = 1 mL), calculated from the vial size and BAC water you select. For a blend, set the vial size to the total of all peptides in the vial; each dose is the combined amount, so one unit figure draws every component together in its fixed ratio. Always confirm against your prescription and your syringe markings.

Potential Benefits and Side Effects

Effects reported in published research. Not a promise of results, and not medical advice.

Reported benefits
  • Reported for the components and for the GHRH-plus-secretagogue principle generally; no benefit has been demonstrated for this specific blend in a published trial.
  • Amplified, pulsatile release of endogenous growth hormone from two complementary receptor mechanisms, larger than either component alone.
  • Downstream IGF-1 elevation.
  • A biomimetic GH pulse pattern that preserves feedback regulation, rather than supplying exogenous growth hormone.
  • Ipamorelin's clean profile relative to older secretagogues (no meaningful cortisol, prolactin, or appetite effect).
  • Convenience of a single injection rather than two.
Reported side effects
  • Human adverse-effect data for the blend do not exist; the following are component and class effects.
  • Water retention and edema.
  • Joint pain and tingling or carpal tunnel symptoms.
  • Head-flush, mild headache, or lightheadedness shortly after injection.
  • Injection-site reactions.
  • Potential glucose elevation and insulin resistance with sustained GH elevation; longer-term effects are not well characterized.

Warnings and contraindications

Important safety information. This is not exhaustive — read the full prescribing information and consult your prescriber or pharmacist.

Warnings
  • No FDA-approved labeling exists; the following reflect the blend's status and class effects, not established label warnings.
  • This is a blend, not a studied compound. No published clinical trial has evaluated this combination as a blend; whatever evidence exists belongs to the individual components studied separately, and combining them does not carry that evidence forward.
  • The ratio is fixed by the seller, not derived from clinical data. Because all components share one vial, a single draw delivers all of them together and none can be titrated, paused, or dose-adjusted independently. If one component causes a problem, the only options are stopping everything or continuing everything.
  • Pre-blending does not change the regulatory status of the individual components; each substance retains its own status, and the blend as a product is not on any FDA list.
  • Co-lyophilized multi-peptide stability is not established. Compatibility, pH interactions, and per-component degradation rates in a shared vial are generally not characterized by the vendors selling these blends.
  • Raising growth hormone and IGF-1 carries class concerns, including glucose intolerance and insulin resistance, fluid retention and edema, joint pain, and carpal tunnel symptoms. Combining two secretagogue mechanisms amplifies the GH signal and therefore these effects.
  • Sustained IGF-1 elevation raises theoretical concerns about promotion of existing malignancy.
  • Both components are prohibited in sport under the WADA Prohibited List (S2). This blend is not usable by tested athletes.
  • Component status: both CJC-1295 and Ipamorelin were named in the February 2026 HHS announcement of peptides slated to return to Category 1, but Ipamorelin's status has been reported as less settled than others and handled differently across the 503A and 503B pathways. Neither appears on the published July 2026 advisory committee agenda.
  • Do not confuse the no-DAC component with CJC-1295 with DAC, which is long-acting (half-life 6 to 8 days) and produces sustained rather than pulsatile GH elevation; a blend built on the DAC version is a pharmacologically different product.
  • Research-grade material varies in identity and purity; safety in pregnancy and breastfeeding has not been established.
Contraindications
  • Not established; no FDA-approved labeling. Precautionary considerations carry over from the components: active or suspected malignancy (GH and IGF-1 biology), elevated baseline IGF-1 or acromegaly, known hypersensitivity, and use by athletes subject to anti-doping rules.

Reconstitution Steps

How to prepare the lyophilized vial. Confirm specifics with your pharmacist or prescriber.

  1. This is a multi-component blend supplied as a single co-lyophilized vial, so all components share one diluent volume and are drawn together in a fixed ratio. Confirm the total vial mass AND the per-component split before starting; the ratio cannot be adjusted after reconstitution.
  2. For this blend specifically: confirm whether your vial is 10 mg total (5 mg of each) or 20 mg total (10 mg of each), and note that both components are present at equal mass in the common presentations.
  3. Wash your hands and gather supplies: the blend vial, the diluent (typically bacteriostatic water), an alcohol swab, and a sterile syringe.
  4. Let the vial and diluent reach room temperature if they were refrigerated or frozen.
  5. Wipe the rubber stopper of both the blend vial and the diluent vial with a fresh alcohol swab and let them air dry.
  6. Draw the intended diluent volume into the syringe.
  7. Insert the needle into the vial and slowly release the diluent down the inside wall rather than directly onto the powder, to avoid foaming.
  8. Do not shake. Gently swirl or roll the vial until fully dissolved.
  9. Inspect the solution: it should be clear with no visible particles. Do not use if cloudy or containing particles.
  10. Calculate concentration per component, not just for the blend: divide EACH component's mass by the same diluent volume. The calculator on this page computes a single concentration, so run it once per component using that component's mass.
  11. Label the vial with the reconstitution date and the full per-component concentration breakdown, and store as directed under reconstituted storage.

Development: This blend has no development history of its own; no published trial has evaluated the fixed-ratio CJC-1295 no-DAC plus Ipamorelin combination in humans. Its rationale rests on well-established endocrinology: GHRH analogs and GH secretagogues act on separate pituitary receptors and are known to synergize, a principle demonstrated across the class rather than for this product. The component histories differ substantially in quality. The CJC-1295 program (ConjuChem) centered on the long-acting DAC version, and the human pharmacokinetic and safety trials (Teichman 2006; Ionescu and Frohman 2006) were conducted on that form, not the no-DAC form used here; the no-DAC compound is the same GHRH(1-29) backbone without albumin binding and has no dedicated late-stage program. Ipamorelin was developed by Novo Nordisk (NNC 26-0161) and first characterized by Raun and colleagues in 1998 as the first selective growth hormone secretagogue; it reached Phase 2 in humans, primarily for postoperative ileus rather than anti-aging, before the program was discontinued. Neither compound has ever been approved.

Notes: This blend is not FDA-approved, and blends as such are not on any FDA list; compounding eligibility is determined per substance, and pre-blending does not change either component's status. Both components were placed in FDA Category 2 in September 2023 and both were named in the February 27, 2026 HHS announcement of peptides slated to return to Category 1, but neither appears on the published July 23-24, 2026 Pharmacy Compounding Advisory Committee agenda or the announced later-meeting list, and Ipamorelin's status has been reported as less settled than others with 503B outsourcing remaining restricted. Removal from Category 2 is not the same as addition to the 503A bulks list. Verify both components against the current FDA list. Both are prohibited at all times in sport under the WADA Prohibited List (S2). Neither is a controlled substance. (Status current as of mid-2026.)

Pharmacokinetics

Molecular weightNot applicable as a single value; this is a two-component blend. CJC-1295 without DAC (Modified GRF 1-29) is approximately 3,368 Da and Ipamorelin is approximately 711.9 Da (CAS 170851-70-4). Vial mass is the sum of the two components, not a molecular weight.
Half-lifeNot a single value, and the mismatch matters: CJC-1295 without DAC is approximately 30 minutes while Ipamorelin is approximately 2 hours. The components clear on different timescales from the same injection, so their overlap is partial by design rather than optimized.
Time to peakGrowth hormone rises rapidly after subcutaneous dosing from both components and returns toward baseline within a few hours.
ClearanceBoth components are cleared by peptidase degradation; neither is albumin-bound. Detailed human clearance parameters are limited.
SequenceNot a single sequence. Component 1: CJC-1295 without DAC, a tetrasubstituted analog of human GHRH(1-29) with substitutions commonly at positions 2 (D-Ala), 8 (Gln), 15 (Ala), and 27 (Leu) for DPP-4 resistance, lacking the drug affinity complex. Component 2: Ipamorelin, the pentapeptide Aib-His-D-2-Nal-D-Phe-Lys-NH2, derived from GHRP-1.

Storage and handling

TemperatureResearch-grade lyophilized blend is typically stored frozen at -20 degrees C long term and refrigerated at 2 to 8 degrees C short term. Not a clinically stored, FDA-approved product.
Light sensitiveProtect from light and moisture; keep the vial sealed.
Shelf lifeLyophilized powder is generally stable for months when frozen; follow the supplier certificate of analysis. There is no FDA-established drug shelf life, and co-lyophilized blend stability is not independently characterized.
Reconstituted storageAfter reconstitution, solutions are generally refrigerated at 2 to 8 degrees C and used within the supplier-indicated period, with freeze-thaw cycles minimized. A blend's in-use dating is governed by whichever component degrades fastest, and that is not established. No FDA-established in-use dating.

Important Notes

Practical considerations for consistency and safety.

  • This is the strongest of the common blends on mechanistic grounds: the two components hit different pituitary receptors (GHRH receptor and ghrelin receptor) and are known to synergize as classes. But the specific fixed-ratio blend has never been tested in a published human trial.
  • The no-DAC component is short-acting and pulsatile (about 30 minutes). Do not confuse it with CJC-1295 WITH DAC, which is long-acting (6 to 8 days) and produces sustained GH elevation; a DAC-based blend is a different product entirely.
  • The human trial data commonly cited for CJC-1295 were generated with the DAC form, not the no-DAC form in this blend.
  • Ipamorelin's selectivity is its distinguishing feature: unlike GHRP-2 and GHRP-6, it does not meaningfully raise cortisol, prolactin, or appetite.
  • The components have mismatched half-lives (about 30 minutes versus about 2 hours), so a single fixed-ratio injection is a compromise.
  • Both are banned in sport (WADA S2).

Lifestyle Factors

  • In research protocols the pairing is typically dosed to align with natural GH pulses, such as before sleep or after exercise, on an empty stomach.
  • Human lifestyle interactions beyond these contexts are not well characterized.
